What Are Proton Pump Inhibitors?

Nexium, Prevacid, and Prilosec are among the most commonly sold PPIs on the market. Prilosec was the first approved by the FDA in 1989, though there are nearly a dozen varieties sold in the U.S. market today. They reduce the amount of acid in the stomach in order to treat the symptoms of certain gastrointestinal disorders. Though typically given in pill or liquid form, PPIs may also be given intravenously in a hospital.

These medicines may be prescribed to patients suffering from acid reflux, erosive esophagitis, Zollinger-Ellison syndrome, or those with gastric and duodenal ulcers. The FDA estimates roughly 20 million people use PPIs in the United States.

The Side Effects of Proton Pump Inhibitors

While common side effects of PPIs include minor issues like abdominal pain, dizziness, and joint pain, among others, users have recently reported more serious risks. After multiple studies confirmed the risk of PPIs, in 2010 the FDA announced a safety risk for users and demanded that manufacturers add warnings to all prescription and over-the-counter medicines that apply. Again, in 2014, the FDA released another warning about PPIs causing acute interstitial nephritis, which can lead to advanced permanent kidney damage or renal failure.

  • Bacterial infections
  • Higher risk of bone fractures
  • Damage to arteries
  • Heart attack or heart failure
  • Kidney disease, kidney failure, or other kidney injuries
  • Stroke
  • Death
